How you guys deal with delivery rates?

Question

I don't know if I have to set up a multiple delivery method for each provider since they have different rates per location.
E.g.:
Fast Courier - location 1 ($10) Fast Courier - location 2 ($70) Fast Courier - location 3 ($50)
Bulk Courier - location 1 ($50) Bulk Courier - location 2 ($80) Bulk Courier - location 3 ($100)
Is that how I should set up the delivery methods? Need your tips and tricks with this.
